Purified protein quantification at A280

Can the concentration of a purified protein of known composition be estimated directly?

Method: With a known extinction coefficient, calculate concentration using A280 and the Beer–Lambert law.

Considerations: Proteins differ in aromatic residues and disulfide bonds, so one universal conversion factor cannot apply to all of them.
